Novel Approaches to Combat Antibiotic Resistance in Gram-Negative Bacteria

Antibiotics are drugs that either kill or slow the growth of bacteria. Under selective pressure, microbes can acquire drug resistance and these pathogenic strains pose a growing public health threat. Therefore, the Seeker desires novel methods to combat antibiotic resistance, particularly in Gram-negative bacteria.

The Seeker is looking for novel approaches to identify new mechanisms for combating Gram-negative bacterial infections. A compelling solution provides a work plan with rationale and desired outcomes, including, but not limited to, microbial strains, assay formats, readouts, and counter assays, if applicable. Both targeted and phenotypic cell-based approaches are welcomed, however, an in vitro biochemical or biophysical approach without activity in living cells is not preferred. Additionally, the Seeker desires proposals addressing insights into the factors limiting small molecule uptake and retention in Gram-negatives (“permeability and efflux”). A method(s) to generate data and improve our ability to predict compound physiochemical properties in Gram-negatives is highly desired. Efflux pump inhibitors are not preferred.

Eligibility Requirements (Criteria)

– The submitted proposal should include the following:

  • Detailed description of the method that can meet the above Solution Requirements. The process must allow for capturing mechanisms in whole, wild-type Gram-negative cells. Include a testable hypothesis and explain how the approach will ensure novel mechanisms for anti-bacterial therapeutics that have high translatability value.
  • In vitro proof-of-concept data (or preliminary data relevant to proof-of-concept) and description of key experiments needed to establish applicability and feasibility. Provide relevant supporting information in the form of literature citations, patents, and/or industry examples, as appropriate.


– The Seeker may wish to partner with the Solver at the conclusion of the Challenge. The solver should describe their expertise and include a statement indicating their interest in this opportunity.

– The proposal should not include any personal identifying information (name, username, company, address, phone, email, personal website, resume, etc.) or any information the Solvers may consider as their intellectual property they do not want to share.
